Kris Commons has withdrawn from the Scotland squad for Wednesday's friendly with Denmark.
The Celtic midfielder is suffering from a foot injury sustained in Sunday's 1-0 victory at Aberdeen.
Full-backs Alan Hutton and Steven Whittaker pulled out of Craig Levein's pool on Sunday.
Cardiff City pair Kevin McNaughton and Don Cowie have been called up as replacements for the game at Hampden Park.
Commons joined the squad at the team hotel on Monday morning but immediately returned to Celtic's Lennoxtown training complex for treatment.
He reported back to the Scotland camp before being ruled out, affecting Levein's plans, but not significantly.
"He's quite quickly establishing himself as one of the key players within the group," explained Levein.
"Somebody else will have to take his place.
"What is hugely encouraging for me is the quality of the group as a whole is such now that the drop off (in quality) isn't significant at all."
Hutton is suffering from a knee injury and was not in the Tottenham Hotspur squad as the London side defeated Athletic Bilboa 2-1 on Saturday.
Whittaker missed Rangers' 3-1 defeat by Chelsea with a toe injury.
He had been sent off after only 17 minutes of Wednesday's Champions League qualifying defeat by Malmo.
The inclusion of McNaughton and Cowie brings up to three the number of Cardiff players in coach Levein's squad.
Scotland captain Kenny Miller scored his first goal for the Welsh side in his competitive debut for the Championship club in Sunday's 1-0 win away to West Ham United following his summer switch from Bursaspor.
